چکیده
Ulcerative colitis (UC) is characterized by chronic colonic inflammation, impaired barrier function and gut bacterial dysbiosis. Anthocyanin-containing potatoes have been shown to maintain the intestinal in colitic mice. However, role of microbiota anti-colitic effects anthocyanin-containing not clear. This study evaluated protective efficacy purple- red-fleshed using a DSS-induced murine model with intact antibiotic-depleted microbiota. Four-week-old mice (C57BL6) were randomized into four groups, receiving standard control diet, or 20% purple-/red-fleshed potato (PP/RP) supplemented diet. A broad-spectrum antibiotic cocktail was used deplete bacteria. After eight weeks, treated 2% DSS their drinking water for five days induce colitis. The polyphenol composition analyzed LC-MS/MS. Intestinal permeability measured FITC-dextran. Colonic myeloperoxidase (MPO) activities determined o-dianisidine dihydrochloride method. fecal short-chain fatty acids (SCFA) content GC-FID. RT-PCR analyze relative gene expression levels cytokines abundance. predominant anthocyanin detected RP pelargonidin (93.13%). Whereas petunidin (75.57%) malvidin (21.05%) two major anthocyanins present PP. Administration antibiotics resulted reduction SCFA. Antibiotic-treated more prone increase permeability. diets alleviated damage, elevation histology score only non-antibiotic alone suppressed elevated MPO activity MUC2 In summary, colitis-associated epithelial damage increased but microbiota-ablated suggests that microbiome essential potatoes.
